Summary
Türkiye, alan ve üretim potansiyelinin yanında, zengin bir elma genetik çeşitliliğine sahiptir. Ancak, farklı isimlendirmelerden ve çeşit içi varyasyonlardan kaynaklanan çeşit karmaşası yaşanmaktadır. Bu nedenle, yetiştiricilikte kullanılmayan bazı eski elma çeşitlerinin kaybolma riski görülmektedir. Ülkemiz bitki gen kaynaklarının tanımlanması amacıyla; meyve türlerindeki tanımlamaların morfolojik özelliklere dayandığı, DNA düzeyinde yürütülen çalışmalar ise sınırlı sayıda olduğu görülmektedir. Bu çalışmada, söz edilen olumsuzlukların azaltılması amacıyla, “Atatürk Bahçe Kültürleri Araştırma Enstitüsü-Yalova” elma koleksiyonundan alınan 35 yerli ve 2 referans elma genotipi, 17 SSR (Simple Sequence Repeats) markör kullanılarak tanımlanmıştır. Elde edilen verilerde benzer ve sinonim genotiplere rastlanmazken, Tavşanbaşı, Tokat, Yaz Elması ve Demir gruplarında homonim durum tespit edilmiştir. CH01d08 lokusu, tanımlama olasılığı en yüksek lokus olarak göze çarparken, genotipler arası en yüksek benzerlik oranı %94 olarak tespit edilmiştir. Ülkemiz elma kaynaklarının SSR düzeyinde tanımlanmasına yönelik, gerçekleştirilen bu çalışma bulguları, ileride yürütülecek diğer çalışmalara ön veri oluşturacaktır. Aynı zamanda, bitki genetiği (genetik haritalama, gen klonlama vb.), ıslahı (kombinasyon ıslahında kullanılacak olan ebeveynlerin genetik benzerliklerinin belirlenmesi, mevcut çeşitlerde ana-baba-hibrid olası ilişkilerinin ortaya çıkarılması vb.) ve yetiştiricilik (bitkilerin tanısı ve DNA düzeyinde kontrol vb.) alanlarında katkılar sağlayacaktır.

Abstract
Turkey has a rich apple genetic diversity besides its area and production potential. However, there is a kind of confusion arising from different naming and variety variations. For this reason, some old apple varieties which are not used in cultivation face the danger of disapperance. However, it seems that characterization of the fruit species are based on the morphological characteristics and the studies carried out at the DNA level remain limited to define the plant gene sources in our country. In this study, 35 domestic and 2 reference apple genotypes from the "Atatürk Horticultural Research Institute-Yalova" apple collection were identified using 17 SSR (Simple SequenceRepeats) markers in order to reduce the mentioned negativity. Homologous status was determined in Tavşanbaşı, Tokat, Yaz Elması and Demir groups while similar and synonymous genotypes were not found in the obtained data. The CH01d08 locus was found to be the locus with the highest probability of identification, while the highest similarity rate between genotypes was found to be 94%. The SSR data obtained from this work will provide preliminary data for future work of apple germplasm. Moreover, the obtained data will contribute to plant genetics, plant breeding (identification of genetic similarities of parents to be used in combination breeding, detection of parent-hybrid possible relationships in existing varieties) and plant growing.
