Summary
Suni mutaşyon oluşturmanın amacı istenen özellik için mtıiasyon oranım artırmaktır. İstenen genetik değişiklikleri elde etmek için, nıutagenlerin seçimi bir dereceye kadar onların yararlanabiiıriiğme ve etkinliklerine bağlıdır. Çeşitler muıagenik uygu i amal ara farklı tepkiler gösterirler. Mutagenik uygulamalar Mı bitkilerinde bazı fizyolojik zararlara neden olmaktadır. Bu çalışma, Gediz-75 ve Sofu makarnalık buğday (Triticum durum Desf.) çeşitlerinin EMS (Ethyl Methane Sulfonate) uygulaması ve gama ışınlamasına tepkilerini saptamak amacıyla 1996 ve 1997 yıllarında Tokat-Kazova şartlarında yürütülmüştür. Tohumlar 50 Gy, 100 Oy, 150 Oy ve 200 Gy gama ışınları iîe ışınlanmış veya % 0.1. % 0.2, % 0.3 ve % 0.4 EMS ile muamele edilmiştir. EMS uygulaması ön ıslatma yapılmadan 8 saat oda sıcaklığında yapılmış ve uygulama siiıesi sonunda tohumlar 6 saat süreyle yıkanmıştır. Mııtagenlerin uygulandığı çeşitler ayrı ayrı denemeye alınmıştır. Ayrıca, gama ışını ve EMS uygulamaları birbirinden ayrı olarak "Tesadüf Bloklan Deneme Deseninde 3 tekerrürlü kurulmuştur. Muîagenler Mı bitkilerinin araştırılan özellikleri üzerinde farklı etkilere neden olmuşlardır ve bu etkiler çeşitlere göre değişmiştir. Her iki çeşitte de mutagen uygulamaları özellikler üzerinde olumsuz etkiler meydana getirmiştir. Bu olumsu/, etkiler özellikle araştırmada kullanılan yüksek dozlarda (150 Gy. 200 Gy, % 0.3 EMS, % 0.4 EMS) daha belirgin olarak ortaya çıkmıştır. Gama ışınlamasıyla Mı bitkilerinde meydana gelen fizyolojik zarar EMS uygulamasından daha yüksek çıkmıştır.

Abstract
The goal of artificial mutagenesis is to increase the rate of mutation for the desired characteristic. The selection of mutagens depends upon some extent to their availability and effectiveness for achieving the desired genetic changes, Cuitivars have different responses to mutagenic treatments. Mutagenic treatments cause some physiological damages in Mj plant. This study was conducted to determine the responses of Gediz-75 ve Sofu durum wheat cuitivars to EMS treatment and gamma irradiation in Tokat-Kazova conditions in 1996 and 1997. The seeds were irradiated with 50 Gy, 100 Gy, 150 Gy and 200 Gy by gamma rays or treated with 0. i %, 0.2 %, 0.3 % and 0.4 % EMS. The seeds were treated with EMS at 24 °C for 8 hours without presoaked and were washed for 6 hours after treatment. Either of treated varieties were grown separately in the trial. Besides, the treatments of gamma rays and EMS separately were "Randomized Complete Block Design" with three replications. Mutagens caused different effects on investigated characteristics of M] plants and these effects were varied with cuitivars. The mutagen treatments had the negative effects on characteristic in both cuitivars. These effects markedly increased with high doses (150 Gy. 200 Gy, % 0.3 EMS, % 0.4 EMS). The physiological damage with gamma irradiation was higher than EMS treated in M| plants.
